简介
6-氯嘌呤是合成腺嘌呤的主要中间体之一,其经6-位氨化即得到腺嘌呤。腺嘌呤,化学名为6-氨基嘌呤,其衍生物在医药及农用化学品等领域应用广泛。比如,它是合成维生素B4(腺嘌呤磷酸盐)的主要原料;美国Gileadsciences公司开发研制的阿德福韦酯(Adefovirdi-pivoxil,9-[2-[双(特戊酰氧甲氧基)膦酰甲氧基]乙基]腺嘌呤),2002年9月经FDA批准已成为用于治疗乙肝的核苷类似物新药,口服后能有效抑制乙肝病毒DNA链的复制。
制备方法
投料物质的量比为乙酰基次黄嘌呤∶三氯氧磷∶N,N‑二甲基苯胺为1.0∶5.0∶1.0 在装有温度计和机械搅拌的500mL的三口烧瓶内,加入35.6g乙酰基次黄嘌呤(0.20mol)、152153.0g三氯氧磷(1.00mol)、24.2gN,N‑二甲基苯胺(0.20mol),开启搅拌,加热至105℃,保温4h后,蒸除未反应的三氯氧磷。冷却到0℃,加入100mL冰水,用0.1MNaOH溶液调pH值至8。冷却后析出大量黄色固体。过滤,冷水洗涤,干燥,得6‑氯嘌呤27.6g,收率90.0%(以乙酰基次黄嘌呤计,下同。),经检测纯度为99.0%。
生物活性
6-Chloropurine是化学合成的砌块。制备9-烷基嘌呤和6-巯基嘌呤的中间体。 有抗肿瘤活性。
体内研究
6-Chloropurine to S-(6-punnyl)glutathione and further metabolism of S-(6-punnyl)glutathione to 6-mercaptopunne may be involved in the mechanism of the 6-Chloropurine-induced antitumor activity. 6-chloropurine and azaserine have been shown to have synergic antitumor properties in a variety of mouse neoplasms when administered simultaneously.
