简介
2,3,4,6-O-四特戊酰基-alpha-D-溴代吡喃葡萄糖可用于制备卡格列净。卡格列净Canagliflozin是第一个被FDA批准的SGLT2抑制剂药物,能通过将葡萄糖分解后通过肾脏排出体外的方式来降低血糖,用于治疗成人2型糖尿病,具有广阔的前景。
制备方法
以ALPHA-D-葡萄糖为原料,经与特戊酰氯反应保护所有羟基后,再与溴化锌、三甲基溴硅烷在-10℃反应生成中间体四,上保护基的工艺中加入少量氯化亚砜,使特戊酰氯用量降低一半,收率无影响,两步收率都在82%以上,总收率大于67%。
